General Affinity™ SPE Information & Protocols

The following general information and starting protocols apply to most Affinity SPE products.

Affinity, next-generation Solid Phase Extraction (SPE)

GS Labloure ”Affinity” products revolutionize Solid Phase Extraction (SPE). Affinity SPE product range provides greater reproducibility with cleaner, more consistent extracts. Affinity SPE products provide unparalleled performance characteristics compared to liquid-liquid extraction, phospholipid removal and protein precipitation products.

This includes:

higher levels of reproducibility
higher levels of extract cleanliness
reduced solvent requirements
increased sensitivity
greater sample throughput

The proprietary manufacturing process involved in the production of Affinity cartridges and plates, provides an SPE product which eliminates issues normally associated with conventional loose-packed SPE.

AFFINITY product methods

The generic AFFINITY product methods outlined below are designed to be a starting point for most sample extraction protocols. Generic method protocol for GS Labloure’s Affinity SPE cartridges and 96 well plate formats:

Affinity HLB cartridge (Reversed-phase): Condition: 500 μL methanol; Equilibrate: 500 μL water; Load: 100 to 500 μL of sample at 1 ml per minute; Wash 1: 500 μL water; Wash 2: 500 μL 5% methanol in water; Elute: 200 μL - 500 μL methanol

Affinity MCX cartridge (Mixed-mode strong cation exchanger): Condition: 500 μL methanol; Equilibrate: 500 μL water with 1% formic acid; Load: 100 to 500 μL of sample at 1 ml per minute containing 1% formic acid; Wash 1: 500 μL water with 1% formic acid; Wash 2: 500 μL methanol with 1% formic acid; Elute: 200 μL - 500 μL methanol with 2% ammonium hydroxide

Affinity MAX cartridge (Mixed-mode strong anion exchanger): Condition: 500 μL methanol; Equilibrate: 500 μL water with 1% ammonium hydroxide; Load: 100 to 500 μL of sample at 1 ml per minute containing 1% ammonium hydroxide; Wash 1: 500 μL water with 1% ammonium hydroxide; Wash 2: 500 μL methanol with 2% ammonium hydroxide; Elute: 200 μL - 500 μL methanol with 1% formic acid

Affinity WAX cartridge (Mixed-mode weak anion exchanger): Condition: 500 μL methanol; Equilibrate: 500 μL water with 1% formic acid; Load: 100 to 500 μL of sample at 1 ml per minute containing 1% formic acid; Wash 1: 500 μL water with 1% formic acid; Wash 2: 500 μL methanol with 1% formic acid; Elute: 200 μL - 500 μL methanol with 2% ammonium hydroxide

Affinity WCX cartridge (Mixed-mode weak cation exchanger): Condition: 500 μL methanol; Equilibrate: 500 μL water with 1% ammonium hydroxide; Load: 100 to 500 μL of sample at 1 ml per minute containing 1% ammonium hydroxide; Wash 1: 500 μL water with 1% ammonium hydroxide; Wash 2: 500 μL methanol with 1% ammonium hydroxide; Elute: 200 μL - 500 μL methanol with 1% formic acid
